English Abstract
Cellular oxidative stress and disrupted defense mechanisms in the context of environmental toxicants are linked with human infertility as a leading global reproductive health problem. Since mammary carcinogenesis and reproductive dysregulation may involve common mechanisms including DNA damage and impaired signaling of stress-response pathways, we explored the effects of 7,12-dimethylbenz[a]anthracene (DMBA), by itself and in combination with cadmium, zinc or cobalt on fertility along with related biochemical and cellular conditions in female rats. We assessed reproductive success, oxidative stress markers, BRCA1 immunohistochemical expression in mammary tissue, mammary cellular damage and cytotoxicity against MCF-7 breast cancer cells. Female rats were divided into five groups: control, DMBA, DMBA+Cd, DMBA+Zn and DMBA+Co. Fertility was assessed in terms of pregnancy rate, duration of gestation, pup size at birth, stillbirth rates, rates of abortions and sex ratios. Oxidative stress was assessed via levels of MDA, SOD, CAT, and GSH. BRCA1 expression was analyzed through immunohistochemistry in mammary tissue, and cell death was evaluated by apoptosis and necrosis counts. The MTT assay gauged MCF-7 cell viability post-treatment. Results revealed significant reproductive impairments related to treatment, with the lowest pregnancy rate (20%) in the DMBA group versus 100% in controls. With pregnancy rates between 30% and 60%, combined groups had impaired reproductive outcomes, such as smaller pups and elevated stillbirth rates. DMBA+Co produced the highest abortion rate and DMBA+Cd led to a shorter gestation and poor offspring outcome. Biochemical severity of oxidative stress was manifested by a significant increase in MDA and reduced antioxidants (particularly in the DMBA+Cd group, which has shown extreme imbalance). The group with DMBA+Zn had some reduction in oxidative stress, though reproductive function was not completely restored. Altered BRCA1 activity in mammary tissues, suggestive of dysregulated balances of stress responses was shown by immunohistochemistry. Apoptosis and necrotic developments were more present in the mammary tissues of each group events, and showed a significantly higher percentage for the combined DMBA+Co group. (MCF-7) cell survival decreased following treatment in vitro, notably with DMBA+Co and to a lesser extent with DMBA+Zn. In summary, DMBA resulted in severe reproductive, oxidative and mammary tissue changes, cadmium aggravated these effects on the oxidative stress as well as reproduction system induced by DMBA, cobalt presented high cellular apoptosis/cytotoxicity rate and zinc only partially biochemically protective without completely rescuing fertility or tissue integrity. These results highlight the interrelationships of infertility, oxidative stress and BRCA1 mammary response to combined toxic exposures.

ناباروري يك مشكل پزشكي و اجتماعي است كه ريشه در مكانيسمهاي پيچيده دارد. در اين زمينه، عوامل محيطي و ويژگيهاي ژنتيكي مورد بررسي قرار گرفتهاند تا بتوان علت اين وضعيت را پيشبيني كرد. در اين مطالعه، نقش الگوي ژنتيكي ژن BRCA1 در مدل موشهاي ويستار براي پيشبيني تأثير آن بر باروري با القاي سميت در غدد پستاني از طريق ماده 7،12-ديمتيلبنز[a]آنتراسن (DMBA) بررسي شد. علاوه بر اين، اثر تركيبي DMBA با عناصر كمياب شامل كادميوم (Cd)، روي (Zn) و كبالت (Co) نيز مورد مطالعه قرار گرفت. موشها به مدت 20 هفته تحت بررسي قرار گرفتند و سپس سرم موش¬ها جمع¬آوري شده و از نظر ماركرهاي استرس اكسيداتيو شامل مالونديآلدئيد (MDA)، سوپراكسيد ديسموتاز (SOD)، گلوتاتيون (GSH) و كاتالاز (CAT) بررسي شد. همچنين، بافتهاي غدد پستاني جهت بررسي پروتئين BRCA1 با روش ايمونوهيستوشيمي بررسي شدند و ميزان آپوپتوز و نكروز سلولي ارزيابي گرديد. نتايج نشان داد كه تركيب DMBA با Cd و Co تغييرات چشمگيري در سطوح MDA، SOD، GSH و CAT ايجاد كرد. با اين حال، در گروه تيماري شده با DMBA+Zn، تعادل اكسيداسيون و احيا به طور خفيفي بهبود يافت، بهطوريكه تفاوت معنيداري در سطوح SOD و CAT نسبت به گروه كنترل مشاهده نشد، ولي سطح MDA بهطور معنيداري افزايش و سطح GSH كاهش يافت. بيشترين ميزان آپوپتوز و نكروز در بافت غده پستاني در گروه تيمار DMBA+Co مشاهده شد. پايينترين ميزان باروري (33٫33 درصد از موشهاي بارور) در گروه تيمار شده با DMBA+Cd ثبت شد كه با شدت بالاتر استرس اكسيداتيو در اين گروه مطابقت دارد. بنابراين، سميت ناشي از كادميوم ميتواند باعث ناباروري در موشهايي شود كه آسيبهاي شديدي در بافت غدد پستاني نشان ميدهند، كه احتمالاً با اختلال در بيان ژن BRCA1 مرتبط است.
