مقدمه: اختلال عملكرد ميتوكندري از مهمترين عوامل اختلالات شناختي در سالمندي است. شواهد نشان دادهاند كه فعاليت ورزشي ميتواند كيفيت ميتوكندري و عملكرد شناختي را بهبود بخشد. با اينحال، شدت و حجم بهينه آن مشخص نيست. ازاينرو، هدف از پژوهش حاضر، مقايسه تاثير شدت و حجمهاي متفاوت تمرينات ورزشي بر برخي نشانگرهاي كنترل كيفيت ميتوكندريايي و ارتباط آن با عملكرد شناختي در هيپوكمپ رتهاي مدل القاء سالمندي با دي -گالاكتوز بود.
روششناسي: در اين پژوهش 35 سر رت نر 3 ماهه با وزن 20±220 گرم به پنج گروه (7 سر) شامل: كنترل (C)، مدل سالمندي ناشي از دي-گالاكتوز (D-gal)، تمرين تداومي با شدت متوسط + دي-گالاكتوز (MICT+D)، تمرين تناوبي با شدت بالا + دي-گالاكتوز (HIIT+D) و تمرينات تناوبي سرعتي + دي- گالاكتوز (SIT+D) تقسيم شدند. بهمنظور القاء سالمندي، گروههاي D-gal و تمريني به مدت 8 هفته، روزانه (mg/kg)150بهصورت تزريق داخل صفاقي دي- گالاكتوز دريافت كردند. تمرينات ورزشي نيز به مدت 8 هفته، 5 روز در هفته براي گروههاي HIIT+D ( 9 دوره دويدن 5/1 دقيقهاي با شدت Vmax %85 و 9 دوره استراحت فعال 2 دقيقهاي با شدت Vmax %45 (،MICT+D (دويدن با شدت Vmax %60، به مدت 3/33- 5/38 دقيقه) و SIT+D (4-9 دورهي 10 ثانيهاي دويدن سرعتي با شدت Vmax%140- %120 و 4-9 دورهي 60 ثانيهاي دويدن با شدت Vmax%100 ) انجام شد. پس از اتمام مداخلات، يادگيري و حافظه ازطريق آزمونهاي ماز آبي موريس و حافظه اجتنابي غيرفعال و بيان پروتئينها به روش وسترن بلات اندازهگيري شد. دادهها با استفاده از آزمونهاي تحليل واريانس يكراهه و دو راهه و همچنين ضريب همبستگي پيرسون در سطح معناداري 05/0 p ≤ تحليل شدند.
يافتهها: نتايج افزايش معنادار بيان PINK1 و Parkin و كاهش معنادار P62 و LC3II را در گروه D-gal و گروههاي تمريني در مقايسه با گروه C نشان داد (001/0p ≤). از طرفي، هر سه روش تمريني موجب كاهش معنادار PINK1 و Parkin، و افزايش معنادار LC3II و P62 در مقايسه با گروه D-gal گرديدند (001/0p ≤). درحاليكه MICT موثرترين روش در تغييراتPINK1, Parkin و P62 بود، SIT بيشترين افزايش معنادار را در LC3II نشان داد (001/0p ≤). كاهش معناداري در مدت زمان رسيدن به سكو در طي روزهاي يادگيري مشاهده شد (001/0p ≤). فقط درگروههاي HIIT و SIT با D-gal كاهش معناداري در اين شاخص مشاهده شد (05/0≤ p). در شاخص مدت زمان سپري شده در ربع هدف تفاوت معناداري بين گروه C با D-gal مشاهده نشد (05/0≤ p). درحاليكه، افزايش معناداري در گروههاي تمريني با گروه C و D-gal مشاهده گرديد (05/0>p). در ارزيابي حافظه اجتنابي غير فعال، در شاخصهاي: مدت زماني كه طول ميكشد حيوان به ناحيه تاريك برود (STL)، تعداد دفعات ورود حيوان به ناحيه تاريك (NST) و مجموع مدت زمان سپري شده در ناحيه تاريك (TDC)، بين گروه C با D-gal تفاوت معناداري نبود (05/0≤ p). در مقابل، هر سه گروه تمريني افزايش معناداري را در (001/0 ≥p) STL و كاهش معناداري در (01/0 ≥p) NST و (001/0 ≥ p) TDC نشان دادند. تفاوت معناداري بين سه روش تمريني در اين شاخصها مشاهده نگرديد (05/0≤ p). علاوهبراين، هيچ رابطه معناداري بين پروتئينهاي ميتوفاژي با شاخصهاي يادگيري و حافظه وجود نداشت (05/0≤p ).
نتيجهگيري: درمجموع، به نظر ميرسد MICT در تنظيم مثبت بيان پروتئينهاي ميتوفاژي موثرتر است؛ درحاليكه هر سه نوع تمرين موجب بهبود عملكرد شناختي در سالمندي شدهاند.

Comparative Effects of Different Exercise Intensities and Volumes on Mitochondrial Quality Control Markers and Their Relationship with Cognitive Function in the Hippocampus of D-Galactose-Induced Aging Rats Model

Purpose: Mitochondrial dysfunction is a key factor to cognitive impairment in aging. Evidence indicates that exercise can improve the mitochondrial quality and cognitive function; however, its optimal intensity and volume remain unclear. Therefore, the present study aimed to the comparative effect of different intensity and volume of exercise training on some mitochondrial quality control markers and their relationship with cognitive function in the hippocampus of D-galactose-induced aging rat model.
Methods: In this study, thirty five 3 month old male rats (220 ± 20 g) were randomly assigned to five groups (n = 7): control (C), D galactose–induced aging (D-gal), moderate intensity continuous training + D galactose (MICT+D), high intensity interval training + D galactose (HIIT+D), and sprint interval training + D galactose (SIT+D). To induce aging, the D gal and exercise groups received intraperitoneal injections of D galactose (150 mg/kg/day). The HIIT+D protocol consisted of 9 bouts × 1.5 min running at 85% Vmax interspersed with 9 bouts × 2 min active recovery at 45% Vmax. The MICT+D group performed continuous running at 60% Vmax for 33.3–38.5 min. The SIT+D protocol included 4–9 sprint bouts × 10 s at 120–140% Vmax and 4–9 bouts × 60 s active recovery at 100% Vmax. The interventions were performed 5 days per week for 8 weeks. After the interventions, learning and memory were assessed using the Morris water maze and Passive avoidance response tests, and the expression of proteins were measured via Western blotting. Data were analyzed using one way and two way ANOVA, and Pearson correlation, at a significance level of p ≤ 0.05.
Results: The results showed significantly increased expression of PINK1 and Parkin and significantly decreased expression of P62 and LC3II in the D gal compared with the C (p ≤ 0.001). Conversely, all three exercise protocols significantly reduced PINK1 and Parkin expression and significantly increased LC3II and P62 expression compared with the D gal (p ≤ 0.001). While MICT was the most effective protocol in modulating PINK1, Parkin, and P62, SIT produced the greatest increase in LC3II expression (p ≤ 0.001). A significant decrease in escape latency across the learning days was observed (p ≤ 0.001), with significant reductions found only in the HIIT+D and SIT+D compared with the D gal (p ≤ 0.05). No significant differences were found between the C and D gal in time spent in the target quadrant (p ≥ 0.05), whereas significant increases were observed in all exercise groups compared with both the C and D gal groups (p < 0.05). In the passive avoidance test, no significant differences were observed between the C and D gal in step through latency (STL), Number of step-through (NST), Time spent in dark chamber (TDC) (p ≥ 0.05). In contrast, all three exercise groups showed significant increases in STL (p ≤ 0.001) and significant decreases in NST (p ≤ 0.01) and TDC (p ≤ 0.001). No significant differences were found among the three exercise groups for these cognitive indices (p ≥ 0.05). Furthermore, no significant correlations were found between mitophagy related proteins and cognitive functions (p ≥ 0.05).
Conclusions: In summary, MICT appears to be more effective in upregulating the expression of mitophagy related proteins, while all three exercise modalities improved cognitive function in the aging model.
